Affinity selection-mass spectrometry (AS-MS) platform is a biophysical technique for protein-ligand binding study. It is a label-free binding assay technology. The ligands from a chemical library can integrate all binding sites of biological targets, which is useful to discover orthosteric, allosteric, and chemically induced proximity ligands. MS can detect many protein-bound ligands simultaneously, which makes AS-MS unique capability of screening biological targets with chemical mixtures such as small molecules, natural products, peptides, and drug metabolites etc.
